Facing this daunting challenge, an ancient virus derived from sewage \u2014 the <strong>bacteriophage<\/strong> \u2014 is being revitalized by a Shanghai research team, opening a <strong>highly efficient and precise \u201cChinese pathway\u201d<\/strong> to tackle the superbug crisis. Today\u2019s (7th) episode of the <em>Shanghai New Productivity<\/em> series reports: <em>\u201cTargeting Superbugs \u2014 Using Technology to Strengthen the Health Defense Line.\u201d<\/em><\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Bacteriophages are the most abundant microorganisms on Earth. As viruses that specifically attack bacteria, they were explored for antibacterial therapy in multiple countries as early as the last century. However, their use was once stalled due to <strong>inefficient screening, slow matching<\/strong>, and the widespread adoption of antibiotics. Now, as \u201csuperbugs\u201d render antibiotics increasingly powerless, global attention has returned to this \u201cold technology.\u201d<\/p>\n\n\n\n<figure class=\"wp-block-image aligncenter size-full\"><img loading=\"lazy\" decoding=\"async\" width=\"730\" height=\"592\" src=\"https:\/\/www.transplantation.com.cn\/wp-content\/uploads\/2025\/10\/\u4f01\u4e1a\u5fae\u4fe1\u622a\u56fe_68fcdba4-bb4b-4463-9ff1-a88d3e7c3ee4.png\" alt=\"\" class=\"wp-image-7429\" srcset=\"https:\/\/www.transplantation.com.cn\/wp-content\/uploads\/2025\/10\/\u4f01\u4e1a\u5fae\u4fe1\u622a\u56fe_68fcdba4-bb4b-4463-9ff1-a88d3e7c3ee4.png 730w, https:\/\/www.transplantation.com.cn\/wp-content\/uploads\/2025\/10\/\u4f01\u4e1a\u5fae\u4fe1\u622a\u56fe_68fcdba4-bb4b-4463-9ff1-a88d3e7c3ee4-300x243.png 300w\" sizes=\"auto, (max-width: 730px) 100vw, 730px\" \/><\/figure>\n\n\n\n<p class=\"wp-block-paragraph\">But where can we find these \u201clife-saving phages\u201d? The answer may surprise you \u2014 in <strong>sewage<\/strong>. <strong>Professor Zhu Tongyu<\/strong>, Director of the Fudan University Phage Research Institute and academic leader of kidney transplantation at Zhongshan Hospital, smiles as he explains: in their lab refrigerators, there\u2019s no food \u2014 only samples collected from sewage treatment plants across the country. This is their <strong>\u201cphage treasure trove.\u201d<\/strong><\/p>\n\n\n\n<p class=\"wp-block-paragraph\">\u201cAntibiotic development can\u2019t keep up with the speed of bacterial mutation. So nature has gifted us with bacteriophages, the natural enemies of superbugs, which we can often isolate directly from sewage. There\u2019s an interesting phenomenon: if you leave a pool of sewage for two or three weeks, it clears up; the impurities settle, and more importantly, the bacteria are consumed by bacteria-targeting microorganisms \u2014 the phages have eliminated them.\u201d<\/p>\n\n\n\n<figure class=\"wp-block-image aligncenter size-large\"><img loading=\"lazy\" decoding=\"async\" width=\"576\" height=\"1024\" src=\"https:\/\/www.transplantation.com.cn\/wp-content\/uploads\/2025\/10\/95f12cbff69a88541db2b2d905ca2df7-576x1024.png\" alt=\"\" class=\"wp-image-7430\" srcset=\"https:\/\/www.transplantation.com.cn\/wp-content\/uploads\/2025\/10\/95f12cbff69a88541db2b2d905ca2df7-576x1024.png 576w, https:\/\/www.transplantation.com.cn\/wp-content\/uploads\/2025\/10\/95f12cbff69a88541db2b2d905ca2df7-169x300.png 169w, https:\/\/www.transplantation.com.cn\/wp-content\/uploads\/2025\/10\/95f12cbff69a88541db2b2d905ca2df7-768x1365.png 768w, https:\/\/www.transplantation.com.cn\/wp-content\/uploads\/2025\/10\/95f12cbff69a88541db2b2d905ca2df7-864x1536.png 864w, https:\/\/www.transplantation.com.cn\/wp-content\/uploads\/2025\/10\/95f12cbff69a88541db2b2d905ca2df7.png 1080w\" sizes=\"auto, (max-width: 576px) 100vw, 576px\" \/><\/figure>\n\n\n\n<p class=\"wp-block-paragraph\">Using a <strong>unique purification process<\/strong>, Zhu\u2019s team turns what everyone else discards into treasure, capturing <strong>highly effective phages<\/strong>. Even better, phages attack <strong>only bacteria<\/strong> and are harmless to human cells. Side effects are far lower than antibiotics \u2014 among the 200+ patients treated by the team, the incidence of side effects was just 7%, mostly manageable fevers, without the allergic reactions commonly associated with antibiotics.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">One beneficiary is <strong>84-year-old Mr. Wu<\/strong>. His family recalls that after knee replacement surgery, he developed <strong>acute cholecystitis<\/strong> that triggered a <strong>drug-resistant bacterial infection<\/strong>. This overcomes the traditional limitation of requiring physical bacterial samples, significantly improving treatment efficiency. A process that previously took <strong>72 hours<\/strong> \u2014 isolation, screening, and treatment \u2014 can now be compressed to <strong>just one hour<\/strong>, and can handle multiple drug-resistant infections from different patient sites.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">\u201cNow we can use epidemiological information, the time and location where the patient acquired the bacteria, and its characteristics and drug-sensitivity profile to predict its specific strain via our database. Our virtual matching system then provides the phage treatment directly. To scale from <strong>\u201cpersonalized rescue\u201d to \u201cmass-producible weapons,\u201d<\/strong> Zhu and his team are using <strong>synthetic biology<\/strong> to continuously engineer phages. They have already secured <strong>multiple patents<\/strong> in key areas, including formulation development and production processes. With ongoing research, the potential applications of phages continue to expand, promising <strong>entirely new therapeutic pathways<\/strong> for a variety of diseases.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">\u201cPhages do more than just lyse bacteria \u2014 they are also a <strong>research tool<\/strong>. They can serve as <strong>vaccine carriers<\/strong>, be delivered directly to the respiratory tract via sprays, or act as a <strong>tumor therapy delivery platform<\/strong>. Phages are a <strong>platform technology<\/strong> \u2014 not only for treating drug-resistant infections, but also as an important tool for future drug development.\u201d<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">\u201cAfter just <strong>two rounds of phage therapy<\/strong>, the lung infection was under control.
